2. summary
Gentamicin(GEN), one kind of broad-spectrum antibiotic, is effcetive to treat the infectious disease. It has been widely used in different lines of poltry, cattle, agriculture and beekeeping at present. But Gentamicin(CAP) can do harm to human body, it will inhibit the hematopoiesis function of marrow, thereby induce not only the Aplastic Anemia and agranulocytopenia, but also induce disease of human beings’ gastrointesinal tract and affect the nervous system.
Radioimmunologic or gas chromatographic method is applied for Gentamicin residue. Howerver, enzyme immunoassays shows considerable advantages over conventional methods. Radioactive materials are no longer required, and the time is less than the gas chromatographic method.
The Gentamicin Residue Rapid Inspection Device is rapid test for qualitivie detection of Gentamicin in specimens at the sensitivity of 50μg/kg (ppb). The major advantages of the one step test device are that the result can be observed by visual and no additional material is needed.
3. principle
The basis of the test is the antigen-antibody reaction. The one step test device is a competitive immunoassay in which the detector reagent consists of colloidal gold particles coated with affinity purified anti-Gentamicin antibodies. The capture reagent in the assay is a Gentamicin-protein conjugate, which is immobilised on the membrane of the test device. In the test procedure, specimens migrate over the membrane. The more analyte present in the sample, the more effectively it will compete with the Gentamicin immobilised on the membrane for binding to the limited amount of antibodies of the detector reagent. Gentamicin, if present in the specimen above 50μg/kg, will thus prevent the binding of the detector reagent to the Gentamicin immobilised on the membrane, resulting in a positive result.
