1. Introduction
Chloramphenicol (CAP) is a broad spectrum antibiotic. It is commonly used for treating
many kinds of disease of domestic fowl because of its efficiency and low price. However, using chloramphenicol will inhibit the hematogenesis function of bone marrow, moreover, it causes induces aplastic anemia and agranulocytosis. Recently, the residues Chloramphenicol in food threaten the health of human has been severely.
The Chloramphenicol chromatographic diagnostic kit is based on the specific immuno-chemical reactions between antigen and antibody. It relies on the competition between Chloramphenicol residues in the sample and the Chloramphenicol immobilized on T line on the membrane for the Chloramphenicol antibody-dye conjugate. Since the drug has been presenting in the sample, it will compete with the drug immobilized on T line, to the limited amount of dye-antibody. As a sufficient amount of drug in the sample is presenting, the drug will saturate the antibody. And thus, it will show an extremely light or even invisible T line, indicating a positive result. On the other hand, if there is a negative sample (or the amount of the drug is lower than the minimum detectable concentration, 0.3 ppb), it will generate two obvious lines in both the T and C line section.

3. Materials required but not provided
1 Ethyl Acetate
2 n-Hexane
3 0.1 M PBS: dissolve 0.352 g NaH2PO4, 1.099 g Na2HPO4, 8.766 g NaCl and 0.2 g NaN3 in deionized water to 1 L, and adjust pH to 7.4
4. Operation Procedure
4.1 Precaution
4.1.1 Open the package just before use to avoid the test cassette being damped.
4.1.2 Avoid touching the membrane in the cassette.
4.1.3 Every test cassette and dropper is single use only. Please do not reuse.
4.1.4 Ethyl Acetate and n-Hexane are highly flammable, please keep away from fire.
4.1.5 Please use 50 mL conical tube made of polypropylene to prevent corruption during extraction procedures.
4.1.6 Store at 4~30 ℃. Do not freeze.
4.2 Sample preparation
Please read Precaution before use.
4.2.1 Weigh 3±0.05 g homogenized sample(no fat or skin) in a 15 mL tube, add 3 mL distilled water, shake properly.
4.2.2 Add 6 mL Ethyl Acetate and vortex for 10 min. Centrifuge at 4000 rmp for 10 min.
4.2.3 Remove 5 mL of upper layer to a clean glass tube and blow to dryness with nitrogen at 60 ℃.
4.2.4 Dissolve the residue in 0.5 mL n-hexane, then add 0.5 mL 0.1 M PBS (pH 7.4), vortex for 1 min.
4.2.5 Centrifuge at 4000 rmp for 5 min at room temperature.
4.2.6 Discard the upper layer and take 80μL the bottom layer solution for assay.
4.3 Operation procedures
4.3.1 Open a package and take out the test cassette.
4.3.2 Use the provided dropper to obtain the sample. Drop 3 drops (about 80 μL) in the Sample collect region.
Note: Do not drop the sample directly to the test result region. It will affect the result.
4.3.3 Wait for 10-15 min to read the result.
4.4 Test Result Interpretation
4.4.1 Negative (CAP < 0.3 ppb): T line is even or darker than C line.
4.4.2 Positive (CAP > 0.3 ppb): T line is invisible or lighter than C line.
4.4.3 Invalid test: If C line does not show which means the test cassette is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the local distributor.
